ABSTRACT

Oral mucosal drug delivery has the advantages of rapid drug absorption, no first-pass effect and good patient compliance. However, factors such as low drug dissolution, saliva carrying the drug into the gastrointestinal tract and the existence of physiological barriers in the mucosa may affect the mucosal permeation and bioavailability of the drug. Nanotechnology applied to drug oral mucosa delivery can overcome the above disadvantages and obtain efficient absorption effect. This paper describes the physiological structure of oral mucosa and the factors affecting the absorption of drugs in oral mucosa, reviews the application of nanotechnology such as liposomes, solid lipid nanoparticles, nanostructured lipid carriers, nanoemulsions, polymer nanoparticles, polymer micelles and nanohybrid suspensions in oral mucosal drug delivery and the mechanism of promoting drug absorption, summarizes the main problems of current research, and gives an outlook on the application of nano oral mucosal drug delivery system. The main problems of current research are summarized, and the prospects for the application of nano oral mucosal drug delivery systems are discussed.

ABSTRACT

With the aging of population intensifies and the level of population health have improved, thus much attention has been directed to how to delaying or preventing skin aging. Skin aging is associated with age, ultraviolet and lifestyle, mainly characterized as skin sagging, wrinkles, pigmentation, so it is urgent to seek traditional Chinese medicine and related cosmetics to solve the problem of skin aging. Traditional Chinese medicine has the functions of anti-oxidation, enhancing human immunity, promoting body metabolism and regulating endocrine, therefore, it has become a research focus in anti-skin aging. This article reviews the skin aging mechanism and the research advances of traditional Chinese medicine anti-skin aging, in order to provide a reference for future research and development of anti-aging traditional Chinese medicine.

ABSTRACT

Purpose To investigate the effects and mechanism of 17β-estradiol on the apoptosis and inflammation of renal tubular cells in rats with renal ischemia/reperfusion injury. Methods All the female Sprague-Dawley rats were ovariectomized and randomly divided into four groups: Control group, Sham group, I/R group and estrogen plus I/R (E2 + I/R) group (n = 8). Right kidney of the rat was excised and artery of the left kidney was blockaded for 45 min.24 h after the reperfusion, we collected the blood and nephridial tissue of each group. An automatic biochemical analyzer was used to measure the expression level of BUN and Cr in blood. Hematoxylin-eosin (HE) staining was used to observe the pathological changes and the degree of inflammatory reaction of the ischemia/reperfusion injury kidney. Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) method was used to detect the apoptosis of renal tubular cells. The expression levels of Cleaved-Caspase-3 protein were measured by Western blot, while the numbers of CD4+ T lymphocyte infiltration in each group were tested by immunofluorescence (IF). Results Compared with the Sham group, expression level of BUN, Cr and Cleaved-Caspase-3 in I/R group significantly increased (P<0.05) as well as the number of apoptotic cells (P<0.05). In the meantime, inflammatory reaction significantly aggravated (P<0.05) and the number of CD4 + T lymphocytes increased remarkably (P<0.05). However, expression level of BUN, Cr and Gleaved-Caspase-3 in E2 + I/R group decreased significantly (P<0.05) and the pathological damage in the kidney was alleviated (P<0.05) compared with I/R group, furthermore, the number of apoptotic cells decreased (P<0.05) compared with I/R group. The inflammatory reaction significantly blunted (P<0.05) and the infiltration of CD4 + T lymphocytes decreased remarkably (P<0.05) compared with I/R group. Conclusion Estrogen can inhibit the expression of Cleaved-Caspase-3 in renal tissue during ischemia/reperfusion injury and reduce the apoptosis of renal tubular cells. It can also reduce the infiltration of CD4 + T lymphocytes, thus playing a protective role on renal ischemia/reperfusion injury.

ABSTRACT

Drug-drug interactions have become a serious problem in the clinic, since plant-based medicines are extensively used. The present study investigated the effects of Ziziphus jujuba fruit (ZJ) extract on the pharmacokinetics of phenacetin, a typical substrate of a cytochrome P450 enzyme CYP 1A2, in rats. The rats were pretreated with the water extract (1.0 g · kg(-1)) or the ethanolic extract (3.6 g · kg(-1)) of ZJ for 10 days, and the pharmacokinetics of phenacetin was investigated after intravenous administration. In an in vitro assay, acetaminophen formation in the hepatic microsomes of ZJ-treated rats was investigated to assess CYP1A2 activity. Our results demonstrated that the treatment with the water and ethanolic extracts of ZJ decreased the plasma concentration of phenacetin and increased the plasma concentration of acetaminophen, resulting in a 43.2% and 15.5% reduction in the AUC0-120 of phenacetin, respectively, and a 53.2% and 64.9% increase in the AUC0-120 of acetaminophen, respectively after intravenous administration. The water or ethanolic extract of ZJ significantly increased the clearance of phenacetin and acetaminophen formation in hepatic microsomes. In conclusion, ZJ extracts displayed effects on the pharmacokinetics of phenacetin and increased the CYP1A2 activity in rats. Therefore, precaution on drug-drug interactions should be taken when ZJ is co-administered with drugs metabolized by CYP1A2, which may result in decreased concentrations of these drugs.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of dihydroartemisinin on the apoptosis of and the vascular endothelial growth factor (VEGF) expression in prostate cancer cell line PC-3M in androgen-independent prostate cancer.</p><p><b>METHODS</b>PC-3M cells were treated with different doses (0, 25, 50 and 100 micromol/L) of dihydroartemisinin for 48 hours, their growth activity analyzed by MTT colorimetric assay and flow cytometry, and changes in the activities of caspase-3 and -8 detected by colorimetric assay. The expression of VEGF mRNA was determined by semi-quantitative RT-PCR, and that of the VEGF protein by Western blotting.</p><p><b>RESULTS</b>Compared with the 0 micromol/L control group, the 25, 50 and 100 micromol/L dihydroartemisinin groups showed significantly increased apoptosis of PC-3M cells ([2.92 +/- 0.45]% vs [8.85 +/- 0.74]%, [12.83 +/- 0.84]% and [18.65 +/- 1.24]%, P < 0.01), and dose-dependent increase in the activities of caspase-8 ([0.47 +/- 0.05 ] U/microg vs [1.22 +/- 0.15], [1.76 +/- 0.07] and [2.91 +/- 0.24] U/microg, P < 0.01) and caspase-3 ([0.44 +/- 0.07] U/microg vs [0.95 +/- 0.08], [1.48 +/- 0.14] and [2.92 +/- 0.45] U/microg, P < 0.01). The expressions of VEGF mRNA and protein were decreased in a concentration-dependent manner.</p><p><b>CONCLUSION</b>Dihydroartemisinin can significantly suppress the growth of PC-3M cells, promote their apoptosis and reduce the expressions of VEGF mRNA and protein, which may serve to explain its inhibitory effect on tumor and angiogenesis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To improve the clinical diagnosis and treatment of primary non-Hodgkin's lymphoma of male genitalia.</p><p><b>METHODS</b>We retrospectively reviewed the clinical data of 5 cases of primary non-Hodgkin's lymphoma of male genitalia, 4 in the testis and 1 in the penis, we also analyzed the relevant literature and clinical significance of the disease.</p><p><b>RESULTS</b>All the 5 cases were treated by surgery and pathologically confirmed to be non-Hodgkin's lymphoma. Three of them received chemotherapy, and the other 2 (1 in the testis and 1 in the penis) underwent both chemotherapy and radiotherapy after the operation. Follow-up averaged 25 months, during which 1 of the patients died and the other 4 survived.</p><p><b>CONCLUSION</b>Primary non-Hodgkin's lymphoma of male genitalia is an uncommon disease with atypical clinical presentations and poor prognosis, which occurs mostly in elderly males. Definite diagnosis of the disease mainly depends on histopathology and immunohistochemistry. Surgery with multiagent chemotherapy and radiotherapy is advisable for its treatment.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To clarify the anatomical relationship of the structures in the first toe webbing space for better dissection of toes in thumb reconstruction.</p><p><b>METHODS</b>The first dorsal metatarsal artery, the first deep transverse metatarsal ligament and the extensor expansion were observed on 42 adult cadaveric lower extremities. Clinically the method of tracing the first dorsal metatarsal artery around the space of the extensor expansion was used in 36 cases of thumb reconstruction.</p><p><b>RESULTS</b>The distal segments of the first dorsal metatarsal artery of Gilbert types I and II were located superficially to the extensor expansion. The harvesting time of a toe was shortened from 90 minutes to 50 minutes with 100% survival of reconstructed fingers.</p><p><b>CONCLUSIONS</b>The distal segment of the first dorsal metatarsal artery lies constantly at the superficial layer of the extensor expansion. Most of the first metatarsal arteries of Gilbert types I and II can be easily located via the combined sequential and reverse dissection around the space of the extensor expansion.</p>

ABSTRACT

Objective To investigate the relationship between quality of life and liver function in patients with chronic viral hepatitis B.Methods Psychological condition was investigated in 536 patients of chronic viral hepatitis B and 200 normal volunteers with self-administered 36-item short-form health survey questionnaire.These patients were divided into four groups based on their liver fuction normal,mild, moderate and severe damage,based on their serum activity of alanine aminotransferase(ALT)and serum level of bilirubin at the time of investigation.The patients with abnormal liver function at the beginning of the study would be retested when liver function recovered to normal level.Before and after comparison was made for those with abnormal liver function by Wilcoxon signed rank test.Trend test for the changes in liver function was performed by Jonckheere-Terpstra test.Results There was significant difference in physical functioning,role limitation due to physical health,bodily pain,general health perception,vitality,social functioning,role limitation due to emotional problems and mental health between patient with chronic viral hepatitis B and healthy volunteers.It was positively correlated with the degree of liver function damage. Scores of general health perception,role limitation due to physical health,role limitation due to emotional problems and vitality were relatively low in patients with chronic viral hepatitis B.Conclusions Quality of life in patients with chronic viral hepatitis B was far worse than that in healthy normal people,which related with their liver function.The more damage to liver function,the worse quality of life.Additional psychological treatment will be helpful for the patients with chronic viral hepatitis B other than drug treatment.
